Monoclonal antibodies (mAbs) are successful as therapeutics due in part to their ability to induce the destructive capabilities of the immune system against specific target cells. mAbs activate antibody-dependent cellular cytotoxicity (ADCC) by natural killer (NK) cells and antibody-dependent cellular phagocytosis (ADCP) by macrophages, and these activities are considered to be important mechanisms of action for many of these mAbs in vivo. Several mAbs also activate the classical complement pathway and promote complement-dependent cytotoxicity (CDC), although the level of efficacy varies greatly, depending on the mAb, the target antigen, and the tumor type. All of these effector functions are mediated by the Fc region of antibody and over the past decade, engineering of this region to enhance the cytotoxic activity of therapeutic antibodies has been a subject of intense investigation.

Phagocytosis involves a series of events, starting with the recognition and binding of cell surface receptors to the particles, then forming an actin-rich membrane extension around the particle, and finally, the membrane extensions fuses to form phagosomes. Pathogens in the phagosomes are destroyed by lowered pH, hydrolysis, and radical attack. These early events, mediated by the innate immune system, are critical to the survival of the host. As a result of this process, pathogen-derived molecules can be presented at the cell surface (antigen presentation), allowing the induction of acquired immunity.

Alkaline phosphatase (AP) staining is a rapid method for screening pluripotent cells by using AP activity. AP is a hydrolase responsible for the dephosphorylation of many molecules, such as nucleotides, proteins, and alkaloids under alkaline conditions. AP activity is higher in pluripotent cells but is greatly decreased in more differentiated cell types. The technique described herein may be used to enumerate pluripotent cells during differentiation in the presence or absence of specific genetic manipulations or small chemical modulators. It may also be used to monitor induced pluripotency using defined factors from more differentiated cell types.

CD133 (also known as Prominin-1 or AC133) has been identified as an important cell surface marker. Cells expressing the CD133 marker are commonly referred to as primitive hematopoietic stem and progenitor cells (HSCs and HPCs). While cells expressing the CD133 marker are usually undifferentiated embryonic stem cell lines, this cell type is usually harvested from bone marrow and cord blood but can be also found in multiple locations including the liver, kidney, prostate, muscle, and neural tissue.CD133+ cells have stemness properties such as drug-resistance, self-renewal, differentiation ability, high proliferation and they are able also to form tumors in xenografts. These cells with CD133+ are more resistant to radiation and standard chemotherapy than CD133- cells.
